NO regulates intracellular and intracellular Ca2 + changes and cell wall construction during pollen tube growth

Nitric oxide (NO) plays a very important role in the growth and development of plants. Recently, the research team of Lin Jinxing from the Institute of Botany, Chinese Academy of Sciences has deeply studied the regulation of NO on Ca2 +, microfilament skeleton, vesicle transport and cell wall construction during the growth of pollen tubes of gymnosperm P. chinensis.

As an important signaling molecule, NO participates in regulating the polar growth of pollen tubes. Through the application of microinjection, non-invasive micro-testing, immunofluorescence labeling and other technologies, it was found that NO release agents promote pollen germination and pollen tube elongation, and have a concentration effect, while inhibitors inhibit pollen germination and pollen tube growth, while making pollen tubes The apex swells and loses its polarity; the NO release agent promotes extracellular Ca2 + influx, the tip Ca2 + concentration gradient increases, the NO inhibitor inhibits the extracellular Ca2 + influx, and the tip Ca2 + concentration gradient decreases.

In addition, the NO releasing agent promotes vesicle transport and depolymerizes the microfibrils at the top of the pollen tube. The NO inhibitor has the opposite effect. At the same time, NO increases the esterified pectin at the top of the pollen tube and decreases the acidic pectin.

In the pollen tube of Pinus sylvestris, NO promotes the inflow of extracellular Ca2 +, thereby maintaining the intracellular Ca2 + concentration gradient, which in turn affects the assembly of the microfilament skeleton at the top of the pollen tube, promotes vesicle transport, and accumulates the esterified pectin at the top of the pollen tube. Normal growth of pollen tubes. Through Ca2 + flow and cytology experiment results, we comprehensively understood the function of NO in the polar growth of pollen tubes.
