The ELISA kit, also known as the enzyme-linked immunosorbent kit, is a commonly used test device in many biopharmaceutical plants and medical laboratories. The main test method is the enzyme-linked immunosorbent assay. ELISA is a microanalytical technique that combines enzyme-catalyzed amplification with specific antigen-antibody reactions. After the enzyme labeled antigen or antibody, it does not affect the specificity of the antigen-antibody reaction, nor does it change the activity of the enzyme itself. Because ELISA kits are inexpensive, accurate, and have short reaction times, they are suitable for large-scale testing and are often used for food safety testing.

The three main points that ELISA needs to follow:

1. The antigen or antibody can adsorb to the surface of the solid support and maintain its immunological activity;

2. The antigen or antibody can be linked to the enzyme by a covalent bond to form an enzyme conjugate which retains its immunological and enzymatic activity;

3. After the enzyme conjugate is combined with the corresponding antigen or antibody, the presence or absence of an immune reaction can be determined according to the color reaction of the added substrate, and the depth of the color reaction is proportional to the amount of the corresponding antigen or antibody in the sample. A standard curve is drawn based on the depth of the color reaction of the known concentration of the antigen or antibody and the concentration of the antibody or antigen in the unknown sample is calculated accordingly.
